Abstract
The new approaches for systematic phosphoprotein analysis by mass spectrometry (MS) were described. The advancements included enrichment and identification of phosphoserine/phosphothreonine proteins from cells, identification of protein phosphorylation sites by capillary LC-MS/MS and evaluation of LC-MS/MS neutral loss scan on a QTOF instrument for phosphopeptide detection and sequencing. 7 human phosphoproteins were identified using a novel mass spectrometry based proteomics strategy. To increase the selectivity of data dependent MS/MS a novel precursor ion discovery tool was evaluated for real time phosphopeptide detection duting LC-MS/MS.
