N2 - FTIR spectromicroscopy is a label-free, nondestructive analytical technique that can provide spatiotemporal information of biomolecules. Performing FTIR measurements on live cells is challenging due to water's strong IR absorption coupled with a need to provide nutrients in liquid. We developed an open-channel device based on a gold-coated porous membrane that allows long-term, continuous IR measurement of adherent cells; subsequently, we tested this device by observing in real-time a living PC12 cell colony for 6.6 days. This technique is promising for noninvasive studies of mammalian systems, capable of providing insight to disease progression, drug discovery, and even cell regulation and development.
