Fas (also known as CD95) is a prototypical death receptor abundantly expressed in liver cells. Engagement of Fas typically triggers a signaling cascade culminating in cell death by apoptosis. Recent studies, however, have demonstrated that several other cellular responses can be initiated by Fas, including proliferation, migration, inflammation, and necrotic cell death. Alterations in Fas signaling are often associated with liver disease. This chapter reviews the molecular mechanisms and regulation of Fas signaling, and the role of Fas in the pathophysiology of the liver.
