T1 - Depression of complex bioelectric discharges in cerebral tissue cultures by thermolabile complement-dependent serum factors

N1 - Funding Information: Initial electrophysiologic studies of sera from rabbits with experimental allergic encephalomyelitis (EAE) , and humans with multiple sclerosis (MS) showed a reversible block of the complex bioelectric synaptic-network discharges of rodent cerebral and spinal cord tissue cultures (2). Marked depression occurred within 5-20 min after exposure to these sera at concentrations of 10-2.576. Abolition of depression by preheating the sera, and restoration, in some cases,b y addition of fresh (unheated) guinea pig serum (10%) suggested that the effect was complement-dependent (see below). (More direct demonstration of complement-dependence was difficult since many of the sera blocked bioelectric discharges even without addition of guinea pig serum.) Similar depression of complex bioelectric activity of mouse cerebellar explants by MS sera was reported by Lums-den (14), and Cerf and Carels (5) obtained remarkably similar electrophysiologic effects consisting of rapid, reversible, complement-dependent depression of polysynaptic reflex responses in freshly isolated adult frog 1 Supported by research grants NS-06545, NS-06735, and NS-11719 National Institute of Neurological Diseaseasn d Stroke, grants MS-760-A 433 from the National Multiple Sclerosis Society, and Kennedy Scholar Drs.
